MonsterScript™ 1st-Strand cDNA Synthesis Kitは、total cellular/polyadenylated RNAを含むmRNAからfull-length first-strand cDNAを合成できるように最適化されています。合成されたcDNAはPCRやReal-Time PCRのテンプレートなど様々なアプリケーションに使用可能です。

MonsterScript 1st-Strand cDNA Synthesis Kitは、RNase H活性を欠いている逆転写酵素であるMonsterScript™ Reverse Transcriptaseを採用しています。RNase H活性を欠く酵素は、長鎖のcDNAやof first-strand cDNA moleculesのより完全なライブラリーを合成するのに寄与します。15-kb RNA templatesを用いてcDNAを合成できます。

キットには最適化された濃度のdNTPs、Mg⁺² 、betaine^*を含んでいるMonsterScript™ RT PreMixが添付されます。Betaineは、逆転写酵素が反応中に停止するのを減少させ、GC-richの領域などの困難な配列の逆転写反応を改善します。キットはoligo(dT)-containing primerとrandom nonamer primerが含まれています。

MonsterScript™ Reverse Transcriptase was used to reverse transcribe six dilutions (8 x106 to 8 x101 pg) of an RNA transcript into cDNA in 20-μl reactions. Ten microliters of each reverse transcription reaction was then used in 50-μl real-time PCR reactions, in duplicate, using EPICENTRE's TAQurate™ GREEN Real-Time PCR MasterMix. A PCR efficiency of 99.4% and /correlation coefficient of 1.000 was achieved.
