ほとんどの真核細胞のmRNAやウィルスRNA、多くの小さな細胞核RNA、異種細胞核RNAを含む多くの細胞核RNA分子には5'末端には"cap"と呼ばれる構造を保持しています。Tobacco Acid Pyrophosphatase (TAP)はcap構造のtriphoshateの架橋における無水リン酸結合を加水分解し、cap nucleosideと5'末端がリン酸化されたRNA分子を生じさせます。cap構造が取り除かれ、5'末端がリン酸化されたRNAは、T4 RNA Ligaseを使用して水酸化されている3’末端と結合したり、末端ラベリングのためにAPex™ Heat-Labile Alkaline Phosphataseを使用して脱リン酸化できます。 同様に、TAPは原核細胞の転写産物の5’末端におけるtriphosphate groupを消化し、5’末端がリン酸化されたRNA分子を生じます。

Unit Definition

One unit of Tobacco Acid Pyrophosphatase releases 1 nmole of inorganic phosphate from m7GpppG in 30 minutes at 37℃ in 50 mM sodium acetate (pH 5.0), 0.1% β-mercaptoethanol, and 1 mM EDTA.

Storage Buffer

50% glycerol containing 10 mM Tris-HCl (pH 7.5), 0.1 M NaCl, 0.1 mM EDTA, 1 mM DTT, and 0.01% Triton X-100.

TAP 10X Reaction Buffer

0.5 M sodium acetate (pH 6.0), 10 mM EDTA, 1% β-mercaptoethanol, and 0.1% Triton X-100.

Quality Control

TAP is tested in an RNA decapping assay and is free of detectable DNA exo- and endonuclease, RNase, and 5'-phosphatase activities.
